MOLECULAR CHARACTERIZATION AND PATHOGENICITY OF NOVEL MALAYSIAN CHICKEN ASTROVIRUS ISOLATES
DESCRIPTION : Chicken astrovirus (CAstV) has for over a decade been associated with runting stunting syndrome, severe kidney disease and visceral gout, and white chick syndrome. However, knowledge of the molecular characteristics and pathogenicity of the virus in day-old specific pathogen-free (SPF) chicks is scarce. This study focused on the characterization of near-complete genome of three Malaysian CAstV isolates following virus propagation in SPF embryonated chicken eggs and pathogenicity in day-old SPF chicks. The three isolates demonstrated unique features including a point mutation in their intergenic regions and an additional stem-loop II-like motif (s2m) in ORF-2. Pairwise sequence comparison and phylogenetic analysis of the ORF-2 amino acid sequence of the three isolates revealed an identity share of 86–91% with group B CAstVs while forming a new subgroup in addition to the known four subgroups (Bi, Bii, Biii and Biv) that exhibit high identity of between 95% and 100% within the subgroups. In the pathogenicity study, birds in the infected and exposed sentinel groups exhibited lethargy and diarrhoea 3 days post-inoculation (dpi) that declined by 6 dpi, and 20% growth retardation by 9 dpi. Mild lymphocytic aggregates in the duodenum, tubular degeneration and interstitial nephritis were observed in the intestines and kidneys, respectively, in both groups. In addition, the mean virus copy numbers of the cloacal swabs were log10 13.23 at 3 dpi and log10 9.04 at 6 dpi for the infected and exposed sentinels, respectively. The study suggests that the Malaysian isolates should be assigned to a new subgroup, Bv within group B CAstV.

DRAFT GENOME SEQUENCE OF MYO-INOSITOL UTILIZING AEROMONAS DHAKENSIS 1P11S3 ISOLATED FROM STRIPED CATFISH (PANGASIANODON HYPOPTHALMUS) IN A LOCAL FISH FARM IN MALAYSIA
DESCRIPTION : A hypervirulent pathotype of A. hydrophila (vAh) is responsible for Motile Aeromonas Septicemia (MAS) and causes mass mortalities among farmed carp and catfish species in the USA and China. One unique phenotype for vAh among other A. hydrophila strains is the ability to utilize myo-inositol as a sole carbon source. While screening for Aeromonas isolates from diseased fish that can grow using myo-inositol as a sole carbon source, A. dhakensis 1P11S3 was isolated from the spleen of striped catfish (Pangasianodon hypopthalmus) displaying clinical MAS symptoms from a freshwater farm in Malaysia. Aeromonas dhakensis is also an important pathogen in aquaculture, and in this study, we report the draft genome sequence for A. dhakensis 1P11S3, that utilize myo-inositol as a sole carbon source.

GENOMIC DATA OF LEPTOSPIRA INTERROGANS HP358 ISOLATED FROM RODENT CAPTURED FROM THE HUMAN LEPTOSPIROSIS SUSPECTED AREAS OF SELANGOR STATE, MALAYSIA
DESCRIPTION : The data provided in this article is the genomic sequence of new Leptospira isolate, Leptospira interrogans strain HP358 (L. interrogans HP358) isolated from rodent, Sundamys muelleri (S. muelleri), captured from the human leptospirosis suspected area, in forest environment, Hulu Perdik, Selangor. The kidney of the rodent was cultured, and the genomic DNA of pure Leptospira isolate was extracted and sequenced. The de novo assembly of genome generated 118 contigs with N50 of 133,176bp. The genome size of the L. interrogans HP358 was determined with a length of 4,808,724 and 35.01% G+C content with 229 subsystems, 5236 coding sequences and 39 RNAs. The whole genome shotgun project has been deposited in NCBI GenBank under the accession number JAFCYY000000000.1.

THE COMPLETE MITOCHONDRIAL GENOMES OF PANGASIUS NASUTUS AND P. CONCHOPHILUS (SILURIFORMES: PANGASIIDAE)
DESCRIPTION : The catfish, Pangasius nasutus and P. conchophilus, are often misidentified between each other due to their similar morphology. Thus, the current study was conducted to differentiate them based on a molecular approach. The complete mitochondrial genomes of P. nasutus and P. conchophilus obtained from the Pahang River (Peninsular Malaysia) were sequenced, assembled, and annotated using Next-generation sequencing (NGS). A 16,465 bp and 16,470 bp length mitogenome sequence of P. nasutus and P. conchophilus, respectively were generated, each containing 13 protein genes, 22 tRNAs, and two rRNAs, typical of most vertebrates. This is the first report of the complete mitochondrial genome sequences of P. nasutus and P. conchophilus. This data is a valuable genetic resource for future studies of these two commercially important species.
