EFFECT OF DIFFERENT DRYING TREATMENTS AND SOLVENT RATIOS ON PHYTOCHEMICAL CONSTITUENTS OF IPOMOEA AQUATICA AND CORRELATION WITH Α-GLUCOSIDASE INHIBITORY ACTIVITY
DESCRIPTION : Ipomoea aquatica is an aquatic plant that is widely consumed in Southeast Asia as a vegetable. In this study, the influence of various ethanol ratios (0, 20, 50, 80, and 100%) as an extraction solvent and different drying methods including air drying (AD), sun drying (SD) and oven drying (OD) on phytochemical constituents of I. aquatica was investigated using a proton nuclear magnetic resonance (1H NMR) based metabolomics approach. The effect on α-glucosidase inhibitory activity and total phenolic content (TPC) was also examined. Clear discrimination was observed between different ethanol ratios and different drying processes by principal component analysis (PCA). The highest α-glucosidase inhibitory activity was observed for absolute ethanol extract from the OD method with IC50 value of 204.0 ± 59.0 µg/mL and TPC value of 22.0 ± 0.7 µg GAE/mg extract. Correlation between the α-glucosidase inhibitory activity and the metabolite were analyzed using a partial least square (PLS) analysis. The metabolites that are responsible for the activity were quercetin derivatives, chlorogenic acid derivatives, sucrose and fructose. This study highlights the basis for future investigations of I. aquatica as a source of food that has the potential for nutraceutical enhancement and as ingredient in medicinal preparation.

URINARY METABOLOMICS STUDY ON THE PROTECTIVE ROLE OF ORTHOSIPHON STAMINEUS IN STREPTOZOTOCIN INDUCED DIABETES MELLITUS IN RATS VIA 1H NMR SPECTROSCOPY
DESCRIPTION : Orthosiphon stamineus (OS) is a herb known in ethnomedicine for treating diabetes mellitus (DM). In this study, a 1H NMR based urine metabolomics tool has been used for the first time to identify the metabolic protective mechanism of OS in DM using Streptozotocin (STZ) induced experimental model in rats. Methods Four different solvent extracts of OS, namely aqueous, ethanolic, 50% aqueous ethanolic and methanolic, at a dose of 500Â mg/kg body weight (bw) were orally administered for 14 days to diabetic rats induced via intraperitoneal injection of 60Â mg/kg bw STZ. NMR metabolomics approach using pattern recognition combined with multivariate statistical analysis was applied in the rat urine to study the resulted metabolic perturbations. Results OS aqueous extract (OSAE) caused a reversal of DM comparable to that of 10Â mg/kg bw glibenclamide. A total of 15 urinary metabolites, which levels changed significantly upon treatment were identified as the biomarkers of OSAE in diabetes. A systematic metabolic pathways analysis identified that OSAE contributed to the antidiabetic activity mainly through regulating the tricarboxylic acid cycle, glycolysis/gluconeogenesis, lipid and amino acid metabolism. Conclusions The results of this study validated the ethnopharmacological use of OS in diabetes and unveiled the biochemical and metabolic mechanisms involved.
